Objective:Non small cell lung cancer (NSCLC) is oneof the most aggressive types of cancer and the leading cause of cancermortality, yet the pathologic mechanisms underlying its devastating clinicaloutcome remain elusive. We focused on identifying the key miRNAs involved inNSCLC prognosis. Method: miRNA expression profiles was analyzed in theformalin-fixed paraffin-embedded specimens from patients with stage IIIA-N2non-small cell lung carcinoma (NSCLC). The prognostic significance of miRNA wasperformed by Kaplan-Meier survival analysis and Cox regression survivalanalysis. The biological functions of miRNA were determined by quantitativereal-time PCR, western blotting, luciferase reporter assay, MTS assay andfluorescent activated cell sorter (FACS) in the cell lines. Result: Patientswith miR-1285-3p down regulation was closely correlated with the poor outcomeof NSCLC (HR=0.4730, 95%CI 0.2855 to 0.7837 for overall survival). Moreover,miR-1285-3p suppressed cell proliferation and enhanced cisplatin sensitivityand radiation sensitivity in non-small cell lung cancer cells via suppressionof the expression of its target gene YAP1 at posttranscription levels. Conclusion:Our results demonstrate that miR-1285-3p play a tumor suppressor role inNSCLC via mediating hippo signaling pathway, and may represent a novelprognostic biomarker and therapeutic target for patients with NSCLC.
